Retatrutide: reading the weight-loss number in a trial paper
Retatrutide trial headlines usually give one weight-loss figure, and the same trial can yield two figures. Five questions help place any figure: which analysis produced it, who was in the trial, how long it ran, whether a journal has published it and which material it describes.
The retatrutide trials guide lists each published result. The retatrutide vs tirzepatide comparison shows why two trials with similar designs still do not make a head-to-head result.
Which analysis produced the figure?
TRIUMPH-2 shows the effect of this choice. The Lancet paper reported a mean change of -18.8% for the retatrutide group with the largest change, using the treatment-regimen estimand. Lilly's news release reported -20.8% for the same group using the efficacy estimand, which represents efficacy had all randomized participants stayed on study intervention without starting prohibited weight management treatments. Who was in the trial?
Placebo results move with the population. In TRIUMPH-1, adults with obesity and no diabetes lost 3.9% on placebo over 80 weeks. In TRIUMPH-2, adults with obesity and type 2 diabetes lost 5.1% on placebo over the same 80 weeks. Sources: PMID 42814954 and PMID 42810372.
How long did the trial run?
Trial length changes what a figure means. The phase 2 obesity trial reported weight change at 24 and 48 weeks, and the figures grew with time in every retatrutide group. Source: 2023 New England Journal of Medicine, PMID 37366315.
Is the figure a paper or a topline release?
TRIUMPH-3 and TRIUMPH-4 have sponsor topline figures and no results paper in PubMed on October 15, 2026, so their numbers come from the sponsor's release alone. Sources: Lilly news release, July 23, 2026 and Lilly news release, December 11, 2025.
Match the material to the paper
A trial result belongs to the molecule that Lilly tested. A vial in a laboratory is documented by its own lot report. The Reta 10mg Kit listing at Curo identifies a supplier's material, and Curo's lot-certificate library and Curo's testing-method page describe how lot reports are organized.
